Oh yeah they can make or break your job opportunity. A really good portfolio can get you a job even if the company doesn't have any openings.

Employers have to view a lot of work and inerview a lot of people, so too many pieces and their eyes glaze over and it all runs together, too few and they don't get a good feel for what you can do.

They want as new as possible BUT they will be more impressed with awesome work than how new something is.

You need to tailor your work to reflect they type of company you are applying to. For example if you were applying to ad agency with a real conservative outlook and policy you probably wouldn't want to have pieces that promote extreme gothic fashions in a very out there design.
